Abstract

Approximately 20% of all deaths worldwide may be attributed to cardiovascular disease, and atherosclerosis is the leading cause of these deaths. Atherosclerosis is an inflammatory disease that leads to lipid accumulation in the blood vessel, and molecular changes in the endothelial and vascular smooth muscle cells, leading to fatty streak formation and vessel narrowing by the plaque growth. In atherosclerosis, the lesion develops slowly in the vessel wall and its rupture may cause a sudden heart attack or stroke. Early diagnosis is essential to prevent these complications. Intercellular communication is important in the early stages of cardiovascular diseases, and exosomes play a crucial role in this type of communication. Exosomes are vesicles produced by cells that carry proteins and genetic material; mRNA, small RNA (miRNA and lncRNA), protein, and DNA, from the donor cell to the destination cells, where they have been reported to have diverse effects. Exosome-mediated cell-to-cell communication has been shown to influence the development of atherosclerosis; therefore, it is essential to investigate the mechanism of exosome-mediated intercellular crosstalk in this disease, which may have diagnostic and therapeutic applications. This thesis aims to characterize exosomes formed by the interacting of vascular smooth muscle cells (VSMC) and endothelial cells (EC) in different co-culture models. Novel miRNAs in exosomes formed in such models hold promise as new biomarkers for the early detection of atherosclerosis. Our study utilized three culture models: a direct co-culture model, an indirect culture, and an isolated exosome treatment model. We characterized the exosomes using nanoparticle tracking analysis (NTA) and Western blot experiments. Then we isolated miRNA, from these exosomes and sequenced their RNA to identify potential miRNA markers. The results pointed to hsa-miR-335-5p as a potential candidate. We plan further research to explore the functional role of hsa-miR-335-5p in atherosclerosis.
Tüm dünyadaki ƶlümlerin yaklaşık %20'si kardiyovasküler hastalıklara atfedilebilir ve ateroskleroz bu ƶlümlerin ƶnde gelen nedenidir. Ateroskleroz, kan damarında lipid birikimine, endotel ve damar düz kas hücrelerinde moleküler değişikliklere yol aƧan, yağlı Ƨizgi oluşumuna ve plak büyümesi ile damar daralmasına neden olan enflamatuar bir hastalıktır. Aterosklerozda lezyon damar duvarında yavaşça gelişir ve yırtılması ani bir kalp krizi veya felce neden olabilir. Bu komplikasyonları ƶnlemek iƧin erken tanı şarttır. Hücreler arası iletişim kardiyovasküler hastalıkların erken evrelerinde ƶnemlidir ve eksozomlar bu tür iletişimde Ƨok ƶnemli bir rol oynar. Eksozomlar, proteinleri ve genetik materyali; mRNA, küçük RNA (miRNA ve lncRNA), protein ve DNA'yı donƶr hücreden hedef hücrelere taşıyan hücreler tarafından üretilen ve Ƨeşitli etkilere sahip oldukları bildirilen veziküllerdir. Eksozomları nanopartikül izleme analizi (NTA) ve Western blot deneyleri kullanarak karakterize ettik. Daha sonra bu eksozomlardan miRNA izole ettik ve potansiyel miRNA belirteƧlerini tanımlamak iƧin RNA'larını sıraladık. SonuƧlar potansiyel bir aday olarak hsa-miR-335-5p'ye işaret etti. Aterosklerozda hsa-miR-335-5p'nin işlevsel rolünü keşfetmek iƧin daha fazla araştırma planlıyoruz.
