Abstract

Microphysiological platforms, commonly referred to as organ-on-chip (OoCs) technology, revolutionize biomedical research by integrating microfluidics to mimic human organ environments. Despite significant progress in replicating in vitro conditions, they fall short of fully replicating the complexity of the human physiological environment. Addressing this, incorporating advanced fabrication technologies, such as polydimethylsiloxane (PDMS) micromechanical valves, enhances organ simulations on the device. This integration facilitates meticulous control over fluidic channels, encompassing parameters such as flow rate, pressure, and volume, thereby essential for maintaining OoCs. Due to their numerous advantages, including controllability, high parallelization, full automation, and increased sample and reaction throughput, these devices can be denoted as Smart Cell Culture Systems. This study comprises distinct sections. The initial part of the thesis is focused on integrating pneumatic valves into microfluidic devices, aiming to establish smart cell culture systems. Subsequently, the developed smart cell culture systems are employed for the on-chip generation of a human multiniche bone marrow microenvironment. The third and fourth sections are specifically oriented towards enhancing conventional nanoplasmonic biosensors, with the goal of achieving objective- and label-free detection. This transformation is envisioned to facilitate the transition of these devices into clinical applications for medical diagnostics. To conclude, we have developed nanoplasmonic biosensor integrated smart cell culture systems with a particular focus on multiniche human bone marrow studies. Additionally, we have developed a user-friendly and readily deployable point-of-care (POC) device designed to mitigate neurocognitive decline in patients with metastatic brain tumours. The device enables the detection of clinically significant biomarkers from patients' bodily fluids, especially plasma serum. The limit of detection (LOD) for our device has been determined as femtomolar (fM) levels.
Ƈip üstü organ (OoCs) teknolojisi olarak adlandırılan mikrofizyolojik platformlar, insan organ ortamlarını taklit etmek iƧin mikroakışkanları entegre ederek biyomedikal araştırmalarda devrim yaratmaktadır. İn vitro koşulların taklit edilmesinde ƶnemli ilerlemeler kaydedilmesine rağmen, insan fizyolojik ortamının karmaşıklığını tam olarak taklit etmekte yetersiz kalmaktadırlar. Bunu sebeple, polidimetilsiloksan (PDMS) mikromekanik valfler gibi gelişmiş fabrikasyon teknolojilerinin dahil edilmesi, cihazların organ simülasyonlarını geliştirir. Bu entegrasyon, akış hızı, basınƧ ve hacim gibi parametreleri kapsayan titiz bir kontrolü sağladığından, OoC'lerin işlevi iƧin gereklidir. Kontrol edilebilirlik, yüksek paralelleştirme, tam otomasyon ve artan numune ve reaksiyon verimi gibi sayısız avantajları nedeniyle bu cihazlar Akıllı Hücre Kültürü Sistemleri olarak adlandırılabilir. Bu Ƨalışma farklı bƶlümlerden oluşmaktadır. Tezin ilk bƶlümü, akıllı hücre kültürü sistemleri kurmayı amaƧlayan pnƶmatik valflerin mikroakışkan cihazlara entegre edilmesine odaklanmıştır. Daha sonra, geliştirilen akıllı hücre kültürü sistemleri, insan kemik iliği nişleri mikro ortamının Ƨip üzerinde oluşturulması iƧin kullanılmaktadır. Üçüncü ve dƶrdüncü bƶlümler ƶzellikle geleneksel nanoplazmonik biyosensƶrlerin geliştirilmesine yƶnelik olup, hedefe yƶnelik ve etiketsiz tespit elde etmeyi amaƧlamaktadır. Bu dƶnüşümün, bu POC cihazlarının tıbbi teşhis iƧin klinik uygulamalara geƧişini kolaylaştırması ƶngƶrülmektedir. SonuƧ olarak, ƶzellikle insan kemik iliği nişleri Ƨalışmalarına odaklanarak nanoplazmonik biyosensƶr entegre akıllı hücre kültürü sistemleri geliştirdik. Ayrıca, metastatik beyin tümƶrü olan hastalarda nƶrobilişsel gerilemeyi azaltmak iƧin tasarlanmış, kullanıcı dostu ve kolayca kullanılabilen bir POC cihazı geliştirdik. Cihaz, hastaların vücut sıvılarından, ƶzellikle de plazma serumundan klinik olarak ƶnemli biyobelirteƧlerin tespit edilmesini sağlar. Cihazımız iƧin tespit sınırı (LOD) femtomolar (fM) seviyeler olarak belirlenmiştir.
